RESUMO: A temperatura afeta o desenvolvimento das plantas, portanto o filocrono tem sido usado para prever eventos de desenvolvimento e estabelecer o momento adequado para práticas agrícolas. Esse estudo buscou avaliar mudanças em filocrono e soma térmica até o florescimento de genótipos de aveia desenvolvidos em diferentes décadas em três regimes térmicos; o efeito de altas temperaturas no desenvolvimento de fitômeros; e identificar o estágio de desenvolvimento em que ocorre a transição do meristema para a fase reprodutiva. Três ambientes foram estabelecidos pela semeadura no outono, na primavera e em temperatura constante (17oC), em fotoperíodo indutor. Apesar da alteração dos valores numéricos, houve pouca diferença entre os filocronos dos genótipos. A adição das temperaturas ótima e máxima específicas no cálculo de graus dias demonstrou a estabilidade de filocrono entre ambientes. O ciclo e a soma térmica correlacionaram-se com o número de fitômeros. UFRGS 078030-2 floresceu mais cedo, teve menos fitômeros e maior tolerância a altas temperaturas do que os demais genótipos. Os genótipos mais recentes transitam para a fase reprodutiva em estágios mais precoces da escala Haun, em comparação com os genótipos mais antigos.

ABSTRACT: Temperature affects plant development therefore phyllochron has been used as a predictor for developmental events to define the time for agricultural managing practices. This study aims to evaluate changes in phyllochron and thermal sum required for flowering by oat genotypes developed at different decades at three temperature regimes; the effect of high temperature on phytomere development; and identify the development stage at the moment of meristem transition to reproductive stage. Three environments were obtained by sowing in the fall, in the spring, and under constant temperature (17oC), always at inductive photoperiod. Despite changes in nominal values small differences were found among genotypes’ phyllochron. Adding specific optimal and maximum temperatures into the growing degree days’ calculation demonstrated phyllochron stability among environments. Plant cycle length and thermal sum correlated with the number of developed phytomeres. UFRGS 078030-2 plants flowered earlier, had a small number of phytomeres, and greater tolerance to elevated temperatures than the other genotypes. More recent genotypes transit to reproductive stage at an earlier Haun stage than older ones.

RESUMO: A evolução da virulência de Puccinia coronata f. sp. avenae desafia a resistência genética de cultivares de aveia à ferrugem da folha. Novas fontes de resistência são constantemente necessárias para estabilizar a produção e reduzir custos. Os objetivos desse estudo foram caracterizar a resistência de seis genótipos de aveia à ferrugem da folha e avaliar o impacto produtivo e econômico causado pela doença. Foram conduzidos ensaios em dois ambientes com e sem o controle químico. A resistência foi medida via progresso da doença, taxa aparente de infecção, severidade final e tamanho da pústula. As perdas foram avaliadas em rendimento, massa de mil grãos e massa do hectolitro. UFRGS 16Q6030-2 foi imune. Em ambos os ambientes, UFRGS 166091-2 e URS Brava exibiram os maiores níveis de resistência e as menores reduções no rendimento de grãos, enquanto URS 22 teve a maior suscetibilidade, com redução superior a 70% no rendimento de grãos. Os resultados obtidos superam os impactos negativos da doença previamente relatados em aveia, particularmente em relação ao rendimento de grãos. Os dados apresentados neste trabalho ressaltam a importância da resistência genética, particularmente a parcial, na manutenção do potencial genético produtivo, na redução de contaminações ambientais pelo menor uso de fungicidas e na ampliação dos ganhos econômicos com a cultura da aveia.

ABSTRACT: The evolution in virulence of Puccinia coronata f. sp. avenae has challenged the genetic resistance to crown rust in oat genotypes. New resistance sources are constantly required for yield stabilization and costs reduction. This study aimed to characterize the crown rust resistance of oat genotypes and the disease´s productive and economic impact. Experiments were conducted in two environments, with and without chemical control. Resistance was measured by disease progress, apparent rate of infection, final severity and pustule size. Damages were based on reduction of yield, 1000 grain mass and hectoliter mass. UFRGS16Q6030-2 was immune. In both environments, UFRGS166091-2 and URS Brava exhibited the greatest level of resistance and the smallest grain yield reduction, while URS 22 was highly susceptible, reducing more than 70% of its grain yield. These results surpass the negative impacts of the disease previously reported in oats, particularly related to grain yield. The data presented in this paper highlighted the importance of genetic resistance, particularly partial resistance, for maintaining oat genetic yield potential, reducing environmental contamination with less fungicides, and increasing economic gains with oat cultivation.

RESUMO A cultura do tabaco tem enfrentado crescentes problemas com doenças nas lavouras do sul do Brasil nos últimos anos. Dentre elas, o mofo branco, causada por Sclerotinia sclerotiorum, tem se destacado. O perfil genético do patógeno ou de seus níveis de agressividade, assim como de resistência em genótipos de tabaco, são ainda escassos. Assim, avaliou-se o perfil genético de 33 isolados de S. sclerotiorum de diferentes municípios dos estados do Rio Grande do Sul, Santa Catarina, Paraná e São Paulo, e da agressividade de 10 isolados em cinco genótipos de tabaco. A agressividade foi testada em casa de vegetação, inoculando-se micélio do fungo em hastes de plantas de tabaco mediante perfuração com palito de dente estéril. O perfil genético dos 33 isolados foi avaliado pela técnica de microssatélites. Isolados do fungo e genótipos de tabaco apresentaram diferentes perfis de agressividade e resistência, respectivamente. Foram detectados 114 alelos com média de 11 alelos por locus e clones não foram observados. Alguns marcadores apresentaram alelo nulo em alguns genótipos, em especial o marcador 99, o qual foi nulo em 14 isolados. Alelos exclusivos foram observados para 61% dos isolados. A análise da informação do conteúdo de polimorfismo (PIC) foi altamente informativa para todos os marcadores. Os isolados provenientes de diferentes Estados do Sul do Brasil não formaram grupos distintos, indicando que o local não foi um fator determinante da variabilidade. A análise de Cluster indicou que as populações menores, originárias do Paraná e Rio Grande do Sul não diferem geneticamente da população maior de Santa Catarina.

ABSTRACT Tobacco has faced rising problems due to diseases in Southern Brazil in recent years. Among the main diseases, the white mold caused by Sclerotinia sclerotiorum has significant impact. Information about the genetic profile of the pathogen, its level of aggressiveness, as well as the resistance of tobacco genotypes is still scarce. Therefore, in this study, we evaluated the genetic profile of 33 isolates of S. sclerotiorum from different places in the states of Rio Grande do Sul, Santa Catarina, Paraná and São Paulo, and the aggressiveness of 10 isolates in five tobacco genotypes. Aggressiveness was tested by inoculating fungus celium on stalks of tobacco plants in a greenhouse. The genetic profile of the 33 isolates was assessed by microsatellite technique. Fungus isolates and tobacco genotypes showed different profiles of aggressiveness and resistance, respectively. A total of 114 alleles were detected with an average of 11 alleles per locus and no clones were observed. Some markers showed null allele in some genotypes, particularly marker 99, which was null in 14 isolates. Unique alleles were found for 61% of the isolates. The analysis of polymorphism information content (PIC) was highly informative for all markers. Isolates from different States of Southern Brazil did not form distinct groups, indicating that the origin site was not a determining factor of variability. The Cluster analysis indicated that smaller populations from Paraná and Rio Grande do Sul do not differ genetically from the larger population from Santa Catarina.

RESUMO: A toxicidade do alumínio (Al) às plantas é observada em cerca de 15% dos solos no planeta, sendo um fator restritivo à produtividade em terras cultiváveis. No Brasil, os solos ácidos são limitantes à produção de trigo (Triticum aestivum L.) e outros cereais. O Al é tóxico para a maioria dos cereais de inverno, quando a sua concentração aumenta e o pH do solo atinge valores inferiores a 5. Uma das principais preocupações sobre o solo ácido é o aumento da mobilidade dos íons Al3+. O Al pode se ligar as paredes celulares das raízes e, como consequência, impedir o alongamento meristemático em espécies sensíveis, provocando danos ao sistema radicular, que resulta em menor desempenho agronômico das plantas. O Al3+ é também capaz de formar complexos altamente estáveis com fósforo (P), limitando sua disponibilidade para as plantas, e também reduzindo a divisão e o alongamento celular. Para lidar com a toxicidade ao Al, as plantas desenvolveram algumas estratégias como a exsudação de ácido orgânicos (AOs) pelas raízes, sendo este mecanismo de destoxificação bem caracterizado. Os AOs, por sua vez, quelam ions Al3+ formando compostos não tóxicos que não penetram no sistema radicular. Alguns genes responsáveis pela tolerância ao Al em trigo foram identificados, com ênfase para TaALMT1 e TaMATE1B, que exsudam os AOs malato e citrato, respectivamente. Nesta revisão, discutimos os mecanismos pelos quais Al danifica raízes, bem como plantas protegem-se, através de dois genes principalmente. Também apresentamos a interação do gene ALMT1 com P e ferro (Fe).

ABSTRACT: Aluminum (Al) toxicity in plants is seen in about 15% of the soils worldwide, restraining yields in arable land. In Brazil, acidic soils limit production of wheat (Triticum aestivum L.) and other cereals. Al is toxic for most winter cereals when its concentration increases and soil pH is below 5. One of the main concerns with acidic soil is the increase in the mobility of Al3+ions. Al binds to cell walls in roots, preventing meristematic elongation in sensitive species, causing damage to the root system and results in lower yields. Al3+ forms highly stable complexes with phosphorus (P), limiting its availability to plants, as well as reducing cell division and elongation. To deal with Al toxicity, plants have developed strategies such as organic acid (OA) exudation by roots; this mechanism of detoxification has been well-characterized. OAs, in turn, chelate ions Al3, forming non-toxic compounds that do not penetrate the root system. Some genes responsible for Al tolerance in wheat have been identified, particularly TaALMT1 and TaMATE1B that transport malate and citrate OAs, respectively. In this review, we discussed the mechanisms by which Al damages roots those by which plants are protected, primarily through two genes. We also described the interaction of the ALMT1 gene with P and iron (Fe).

Abstract The barley HvAACT1 gene codes for a citrate transporter associated with tolerance to acidic soil. In this report, we describe a single nucleotide polymorphism (SNP) in the HvAACT1 coding region that was detected as T-1,198 (in genotypes with lower root growth on acidic soil) or G-1,198 (greater root growth) and resulted in a single amino acid change (L/V-172). Molecular dynamic analysis predicted that HvAACT1 proteins with L or V-172 were stable, although the substitution led to structural changes within the protein. To evaluate the effect of the SNP on tolerance to acidic soil, barley accessions were separated into haplotypes based on the presence of a 1 kb insertion in the HvAACT1 promoter and a 21 bp insertion/deletion. These markers and the SNP-1,198 allowed the identification of five haplotypes. Short-term soil experiments showed no difference in root growth for most of the accessions containing the 21 bp insertion and T or G-1,198. In contrast, genotypes showing both the 21 bp deletion and G-1,198, with one of them having the 1 kb insertion, showed greater root growth. These results indicate that the SNP was not advantageous or deleterious when genotypes from the same haplotype were compared. The occurrence of the SNP was highly correlated with the 21 bp insertion/deletion that, together with the 1 kb insertion, explained most of the barley tolerance to acidic soil.

Abstract Barley is a major cereal grown widely and used in several food products, beverage production and animal fodder. Genetic diversity is a key component in breeding programs. We have analyzed the genetic diversity of barley accessions using microsatellite markers. The accessions were composed of wild and domesticated barley representing genotypes from six countries and three breeding programs in Brazil. A total of 280 alleles were detected, 36 unique to Brazilian barley. The marker Bmag120 showed the greatest polymorphism information content (PIC), with the highest mean value found on chromosome three, and the lowest on chromosomes four and six. The wild accessions presented the highest diversity followed by the foreign genotypes. Genetic analysis was performed using Principal Coordinates Analysis, UPGMA clustering, and Bayesian clustering analysis implemented in Structure. All results obtained by the different methods were similar. Loss of genetic diversity has occurred in Brazilian genotypes. The number of alleles detected in genotypes released in 1980s was higher, whereas most of the cultivars released thereafter showed lower PIC and clustered in separate subgroups from the older cultivars. The use of a more diverse panel of genotypes should be considered in order to exploit novel alleles in Brazilian barley breeding programs.

A presença de alumínio (Al) reduz o rendimento em solos ácidos ou em áreas onde o subsolo possui pH abaixo de 5, pois limita o crescimento radicular e, consequentemente, a absorção de água e nutrientes. Genótipos elite de aveia (Avena sativa L.) não selecionados para a tolerância ao Al foram avaliados quanto a essa característica em solução nutritiva. Foi utilizada, como parâmetro de comparação da tolerância ao Al, a média de recrescimento radicular após a exposição ao Al. O recrescimento da raiz principal dos genótipos elite foi comparado com os controles UFRGS17, considerado tolerante e UFRGS930598-6, sensível. Foram avaliadas as linhagens UFRGS057005-1 e UFRGS057022-2, e as cultivares comerciais 'URSGuria', 'URSTorena', 'URSPenca', 'URSGuará', 'URS Charrua', 'URSTarimba', 'URSTaura', 'URSGuapa' e 'URS21'. A amplitude de recrescimento da raiz dentro de cada genótipo foi elevada, sendo a menor de 15mm e a maior de 44mm. As cultivares 'URSCharrua' e 'URSGuapa' demonstraram tolerância superior a 'UFRGS17'. URSTarimba, apesar da média similar a UFRGS17, mostrou distribuição de frequência mais positiva. URSTorena, UFRGS0570005-1 e URSPenca classificaram-se como intermediários, sendo inferiores a UFRGS17. Nenhum dos genótipos elite apresentou médias de recrescimento igual ou inferior às obtidas pelo controle sensível, UFRGS930598-6.

Oat genotypes were evaluated for aluminum (Al) tolerance in hydroponic solution. The ability of growing after being exposed to high Al (root regrowth) was used to evaluate the genotypes. The apical root regrowth for each genotype was compared to the controls: UFRGS17, Al tolerant, and UFRGS930598-6, Al sensitive. Two lines UFRGS057005-1 and UFRGS057022-2 and nine cultivars 'URSGuria', 'URSTorena', 'URSPenca', 'URSGuará', 'URSCharrua', 'URSTarimba', 'URSTaura', 'URSGuapa' and 'URS21' were evaluated. The root regrowth amplitude inside each genotype was high; the smallest one was 15mm and the largest one, 44mm. URSCharrua and URSGuapa showed higher tolerance than UFRGS17. Despite the similar mean to UFRGS17, URSTarimba regrowth frequency had a more positive skewness. URSTorena, UFRGS0570005-1 and URSPenca were classified as intermediate, being inferior to UFRGS17. None of the genotypes showed root regrowth equal or inferior to the sensitive control, UFRGS930598-6.

Os objetivos deste estudo foram determinar os mecanismos da tolerância à deficiência de P em genótipos de trigo contrastantes e avaliar a associação desses mecanismos à expressão diferenciada de genes. Foram avaliadas plântulas das cultivares de trigo Toropi (tolerante à deficiência de P) e Anahuac (sensível). As plântulas foram cultivadas em hidroponia, na ausência ou presença (1,0 mmol L-1) de P, durante três períodos de tempo: 24, 120 e 240 horas. Os teores de fosfato livre (Pi) e P total foram medidos na parte aérea e nas raízes. O delineamento experimental foi em blocos ao acaso com três repetições, cada uma formada por dez plantas. Foi avaliada a expressão relativa dos genes que codificam o transportador de malato TaALMT1 e o fator de transcrição PTF1. Períodos de deficiência de P superiores a dez dias aumentaram a expressão de TaALMT1 somente em 'Toropi'. A expressão de PTF1 foi induzida em ambos os genótipos no início da deficiência, porém manteve-se significativa após dez dias somente em 'Toropi'. A concentração de Pi livre na parte aérea em 'Toropi' foi independente da disponibilidade de P; sob limitação, 'Toropi' favoreceu a manutenção da concentração de Pi na parte aérea. A tolerância à deficiência de P em 'Toropi' nos estádios iniciais de crescimento é devida principalmente à sua capacidade de manter constante a concentração de Pi na parte aérea.

The objectives of this study were to determine low-P tolerance mechanisms in contrasting wheat genotypes and to evaluate the association of these mechanisms to differential gene expression. Wheat seedlings of cultivars Toropi (tolerant to low-P availability) and Anahuac (sensitive) were evaluated. Seedlings were hydroponically grown in the absence or presence of P (1.0 mmol L-1) during three different time periods: 24, 120 and 240 hours. Free phosphate (Pi) and total P contents were measured in shoots and roots. The experiment's design was in randomized blocks with three replicates, each formed by ten plants. The relative expression of genes encoding the malate transporter TaALMT1 and the transcription factor PTF1 was evaluated. Phosphorus starvation beyond ten days increased the expression of TaALMT1 only in 'Toropi'. PTF1's expression was early induced in both genotypes under P starvation, but remained significant after ten days only in 'Toropi'. Shoot Pi concentration in 'Toropi' was independent from P availability; under starvation, 'Toropi' favored the maintenance of shoot Pi concentration. The low-P tolerance of Toropi cultivar at initial growth stages is mainly due to its ability to maintain constant the Pi shoot level.

Arabidopsis thaliana é uma das espécies mais utilizadas na pesquisa científica atualmente. Apesar de não apresentar importância econômica direta, esta espécie é o foco de pesquisas na área da genética, bioquímica e fisiologia. O número de trabalhos publicados sobre a mesma aumentou significativamente após o seqüenciamento de seu genoma. Apesar do grande número de estudos existe ainda muita desinformação sobre qual o seu verdadeiro papel na pesquisa científica de espécies cultivadas e de que maneira o avanço no conhecimento adquirido com A. thaliana pode auxiliar o desenvolvimento de cultivares cada vez mais resistentes, adaptados e produtivos. Os objetivos deste trabalho são discutir as razões do uso da A. thaliana como espécie modelo e a aplicabilidade deste modelo no estudo de espécies cultivadas.

Arabidopsis thaliana has been the species of choice for scientific research. Despite its lack of economic importance, it has been the focus of genetic, biochemical and physiological research worldwide. The number of published articles about arabidopsis has increased substantially after its genome was sequenced, and outgrew the number of articles related to economically important species. Despite the great number of studies involving arabidopsis, there is much disinformation about the actual role of this species in crop scientific research, as well as how the breakthroughs in arabidopsis research may help to develop more adapted and productive crops. This work aims to discuss reasons for using A. thaliana as a model species and the feasibility of this model for crop studies.

O objetivo deste trabalho foi testar uma metodologia para determinar doses ótimas de N a serem aplicadas em cobertura, no estádio V6 do milho, com base em características de solo e de planta, e verificar quais os indicadores mais precisos para a predição da dose de N a ser aplicada. O experimento foi conduzido em Eldorado do Sul, RS, em dois anos agrícolas. Os tratamentos consistiram de cinco doses de N, aplicadas em duas épocas. Além do rendimento de grãos, determinaram-se, no estádio V6, o teor relativo de clorofila na folha, as massas de matéria seca da folha e da planta, o teor e a quantidade de N acumulados na folha e na planta, teores de nitrato, amônio e N mineral no solo. As características de planta foram mais eficientes para a predição da dose de N em cobertura no milho, nos dois anos agrícolas, do que as características de solo, com destaque para massa de matéria seca e N acumulado na planta, seguidas pelo teor relativo de clorofila na folha. É possível desenvolver ou adaptar a metodologia testada, para predição de doses ótimas de N em cobertura em milho, a partir de características de planta e de solo, desde que se possua uma rede de ensaios com uma curva de reposta a esse nutriente.

The objective of this work was to test one methodology to determine nitrogen (N) optimum rate to side dress at V6 corn stage, using soil and plant characteristics, and to verify which are the most precise indicators of N rate application. A field experiment was carried out in Eldorado do Sul, RS, Brazil, in two growing seasons. The treatments were five N rates applied in two timings. The following variables were measured: corn grain yield, leaf relative chlorophyll content, leaf and plant dry weight, N content and accumulated in the leaf, and plant and soil nitrate, ammonium and N mineral contents were measured at V6. From the evaluated characteristics, the most precise to predict optimum N rate for side dressing in corn were plant dry weight, N accumulated in the plant and leaf relative chlorophyll content. The results show that is possible to adapt or to develop a method to predict the optimum N rates for side dressing, using soil and plant characteristics, since there is a N response curve available from a group of experiments.

O arroz vermelho é considerado a principal planta daninha na maioria das lavouras de arroz, pois, além dos prejuízos causados pela competição, sua similaridade com o arroz cultivado dificulta seu controle. A dormência e a viabilidade de suas sementes no solo permitem a continuidade dos biotipos na lavoura e facilitam a disseminação para áreas próximas. A duração e a intensidade da dormência variam com o biotipo e com as condições ambientais vigentes durante o desenvolvimento da semente. A maior parte dos ecotipos apresenta dois tipos de dormência, uma devido às estruturas envoltórias e outra devido ao embrião. O período necessário para a superação da dormência é dependente das condições (temperatura e umidade) a que a semente é exposta após a maturação. Várias substâncias têm sido eficientes na quebra da dormência, em especial, ácidos e citocininas. Outros reguladores tiveram apenas efeito parcial. Ressalta-se que todas as substâncias testadas não atuaram sobre as estruturas envoltórias. O mecanismo pelo qual ocorre a manutenção/alívio da dormência permanece desconhecido.

Red rice is considered the major weed in rice areas. The similarity between red rice and cultivated rice difficults the red rice control. Seed dormancy and viability in the soil allow the maintanence of the biotype in the area and facilite the dissemination in surrounded areas. Dormancy duration and intensity depend on biotype and environmental conditions during seed development. In general, biotypes have two types of dormancy: one from surrounding structures and the other one from embryo. The dormancy relief can occur on different times. It depends on environmental conditions (temperature and humidity) in afterripening. Some substances have shown good results on breaking dormancy, in special, acids and cytokinins, however, other hormones have shown only a partial effect. These substances have not affected the surrounding structures. The red rice’s mechanism of dormancy/relief is far from being stabilished.
